Description: The R4-6A2 antibody reacts with mouse interferon-gamma (IFN-gamma ), a homodimeric 15-20 kDa cytokine secreted by Th1 cells. The R4-6A2 antibody is a neutralizing antibody. IFN-gamma is an ∽20 kDa factor produced by activated T, B and NK cells and is an anti-viral and anti-parasitic cytokine. IFN-gamma in synergy with other cytokines, such as TNF-alpha, inhibits proliferation of normal and transformed cells. Immunomodulatory effects of IFN-gamma are exerted on a wide range of cell types expressing the high affinity receptors for IFN-gamma. Glycosylation of IFN-gamma does not affect its biological activity. Applications Reported: The R4-6A2 antibody has been reported for use in immunoblotting (WB), ELISPOT, and ELISA. Applications Tested: The biotinylated R4-6A2 antibody has been tested as the detection antibody in a sandwich ELISA for analysis of mouse IFN gamma (Interferon-gamma, IFN-g) in combination with the purified AN-18 (14-7313) antibody or the purified XMG1.2 (14-7311) antibody for capture and recombinant mouse IFN gamma (39-8311) as the standard. A suitable range of concentrations of this antibody for ELISA detection is 0.5-2 μg/mL. A standard curve consisting of doubling dilutions of the recombinant standard over the range of 200 pg/mL - 1.6 pg/mL (AN-18 assay) or 2000 pg/mL - 15 pg/mL (XMG1.2 assay) should be included in each ELISA plate. Filtration: 0.2 μm post-manufacturing filtered.
