Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous PGA5

  • Recognizes human PGA5
  • Pepsinogen I belongs to the peptidase A1 family
    • Gene encodes 388 amino acid protein with 15 amino acid signal peptide and 46 amino acid propeptide
    • Secreted only by chief cells of corpus and serum level decreases with increasing grade of corpus atrophy
    • Low serum levels of pepsinogen I indicative of chronic atrophy, risk factor for gastric cancer

Process:

  • Antibody specific for PGA5 pre-coated onto microwells
  • PGA5 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for PGA5 is added to detect captured PGA5 protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
