Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous LRRC32.

  • Recognizes human LRRC32
  • LRRC32 (GARP) is a transmembrane protein of 662 amino acids, extracellular portion contains 20 leucine-rich repeats
    • LRRC32 is a cell surface receptor on regulatory T-lymphocytes (Treg cells), platelets, hepatic stellate cells and certain cancer cells
    • Demsonstrated as Treg-specific activation marker
    • Critical for surface expression of latent TGF-β by binding to complex and functioning as cell surface receptor
    • Membrane bound LRRC32 can be shed from cell surface and exist in soluble form

Process:

  • Antibody specific for LRRC32 pre-coated onto microwells
  • LRRC32 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for human LRRC32 is added to detect captured human LRRC32 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
