Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous human

  • Recognizes human FOLR1
  • Folate receptor 1 (FOLR1, folate receptor alpha, adult folate-binding protein, FBP) is a 38 kDa glycoprotein belonging to folate receptor family
    • Secreted protein that anchors to membranes by glycosyl-phosphatidylinsitol linkage or exists in soluble form
    • Expression often limited to apical surfaces of epithelium in lung, kidney and choroid plexus
    • Overexpressed in variety of solid tumors (ovarian cancer, non-small cell lung cancer, breast cancer, kidney cancer, high-grade osteosarcoma)

Process:

  • Antibody specific for human FOLR1 pre-coated onto microwells
  • Human FOLR1 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for human FOLR1 is added to detect captured human FOLR1 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
