Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous ANGPTL4

  • Recognizes human ANGPTL4
  • Angiopoietin-like protein 4 (ANGPTL4) is a member of angiopoietin-like protein subfamily
    • Secretory protein functions as modulator of glucose and lipid metabolism
    • Inhibits lipoprotein lipase (LPL)-dependent lipolysis, limiting uptake of free fatty acids
    • May play role in several cancers and has been shown to prevent metastatic process by inhibiting vascular activity, tumor cell motility and invasiveness
    • Might be biomarker in renal cell carcinoma
    • Decreased expression results in Type 2 diabetes

Process:

  • Antibody specific for ANGPTL4 pre-coated onto microwells
  • ANGPTKK4 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for ANGPTL4 is added to detect captured ANGPTL4 protein
  • For signal development, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
