Gibco™ Geltrex™ Flex LDEV-Free hESC-Qualified Reduced Growth Factor Basement Membrane Matrix
Description
Gibco Geltrex Flex LDEV-Free hESC-Qualified Reduced Growth Factor Basement Membrane Matrix is a soluble form of basement membrane that is purified from Engelbreth-Holm-Swarm (EHS) tumor cells. The major components of Geltrex Flex matrix include laminin, collagen IV, entactin, and heparan sulfate proteoglycan.
Routinely used for attachment and maintenance of human embryonic stem cells (hESCs) and induced pluripotent stem cells (iPSCs), every lot of hESC-qualified Geltrex Flex matrix is quality control tested using hESCs and iPSCs to help ensure support of stem cell culture. For a neural induction of hESC and iPSC protocol, see Induction of Neural Stem Cells from Human Pluripotent User Guide (Pub. No. MAN0008031).
Features of Geltrex Flex LDEV-Free hESC-Qualified Reduced Growth Factor Basement Membrane Matrix
- Each lot tested with iPSCs and H9 hESCs for at least five passages to help ensure reliability and consistency in hESC/iPSC studies
- Each lot tested for the absence of lactose dehydrogenase elevating virus (LDEV), mycoplasma, and a broad panel of viruses and microorganisms
- Lots tested for endotoxin, maintenance of gel stability, and HUVEC tube formation
- Growth factor–reduced to improve lot-to-lot consistency
- Formulated without phenol red to minimize potential for estrogen-like effects
- Provided in recyclable glass vials
- 1-mL, 5-mL, and 10-mL sizes for maximum flexibility. Use only what you need. Reduce need for extensive aliquoting and unwanted freeze-thaws.
For the culture of primary cells, such as epithelial cells, endothelial cells, and smooth muscle cells, and angiogenesis assays, neurite outgrowth assays, or bile canaliculi formation assays with polarized hepatocytes, standard Geltrex Flex matrix can be used.
For the culture of tumoroids and other organoids, Geltrex Flex LDEV-Free Organoid-Qualified Reduced Growth Factor Basement Membrane Matrix is recommended. Each lot of organoid-qualified Geltrex Flex matrix is tested and supports the culture of mouse intestinal organoids, mouse liver ductal organoids, and mouse airway organoids, as well as expansion of mouse small intestinal organoids for more than five passages with typical organoid budding morphology.

Frequently Asked Questions (FAQs)
Both Gibco Vitronectin (VTN-N) Recombinant Human Protein, Truncated (Cat. No. A400457 and A400458) and Gibco Geltrex Flex LDEV-Free hESC-qualified Reduced Growth Factor Basement Membrane Matrix (Cat. No. A4000046801, A4000046802, or A4000046803) have been successfully used with Essential 8 Flex Medium.
We recommend using Gibco Geltrex Flex LDEV-Free hESC-qualified Reduced Growth Factor Basement Membrane Matrix (Cat. No. A4000046801, A4000046802, or A4000046803) or Gibco Vitronectin (VTN-N) Recombinant Human Protein, Truncated (Cat. No. A400457 or A400458) for xeno-free applications.
Yes. We recommend following the coating instructions for Vitronectin or rhLaminin-521 and using the culture and passaging recommendations in the StemFlex protocol.
We have evaluated the use of Cytotune-iPS 2.0 Sendai Reprogramming Kit (Cat. No. A16517) for somatic cell reprogramming of both neonatal and adult human dermal fibroblasts. For fibroblasts, follow the instructions provided in the Cytotune 2.0 reprogramming manual for feeder-free reprogramming (pgs. 16-20). On Day 7, you may use rhVTN-N (Cat. No. A400457 or A400458), Geltrex Flex matrix (Cat. No. A4000046801, A4000046802, or A4000046803), or rhLaminin-521 (Cat. No. A29248 or A29249). From Day 8 onward, rather than feeding daily with Essential 8 Medium, we recommend that you feed reprogrammed fibroblasts every-other-day with StemFlex Medium.
We have evaluated the use of Cytotune-iPS 2.0 Sendai Reprogramming Kit (Cat. No. A16517) for somatic cell reprogramming of CD34+ blood cells. For CD34+ cells, follow the instructions provided in the Cytotune 2.0 reprogramming manual for feeder-free reprogramming (pgs. 39-44). On Day 3, you can utilize rhVTN-N (Cat. No. A400457 or A400458), Geltrex Flex matrix (Cat. No. A4000046801, A4000046802, or A4000046803), or rhLaminin-521 (Cat. No. A29248 or A29249). From Day 8 onward, rather than feeding daily with Essential 8 Medium, reprogrammed CD34+ cells should be fed every-other-day with StemFlex Medium.
