Leucine-rich repeat FLI-I interacting protein 1 (LRRFIP1/FLAP-1) is a MyD88-interacting signal protein which mediates either induction or repression of TLR signaling. Flap-1 directly interacts with β-catenin and activates β-catenin-dependent transcription activity. Flap-1 or LRRFIP1 was also shown to bind the TNF- -308 promoter polymorphism site, suggesting its possible role in the regulation of TNF- production. FLAP-1 along with LRRFIP2 and FLIIH serve as a balance in coordinating inflammatory response after infection, and may act as a novel target in the control of agonist-induced TLR-mediated inflammatory and autoimmune diseases.
